PCR-detection of genetic markers of microorganisms’ antibiotic resistance from milk of the cows at mastitis

Summary. Results of molecular genetic monitoring of the resistance of the main bacterial pathogens isolated from the secretion of the mammary gland of cows during inflammation are presented in the article. Real-time PCR studies were conducted by the authors. The isolated microorganisms included: Staphylococcus spp. (S.epidermidis, S. saprophyticus, S. haemolyticus) - 73,3%, S. aureus - 36,0%, E. coli - 32,0%, S. agalactiae - 6,7% of the number of samples studied. The incidence of Staphylococcus spp. were 25,3%, S. aureus and E. coli in the form of monocultures - 12,0% and 4,0%, respectively. Associations of microorganisms included: S. aureus, Staphylococcus spp. - 14.7%; E. coli, Staphylococcus spp. -21,3%; E. coli, S. agalactiae - 2,7%; S. aureus, Staphylococcus spp. *, E. coli - 8,0%; Staphylococcus spp. *, E. coli, S. agalactiae - 2.7%; S. aureus, Staphylococcus spp. *, S. agalactiae, E. coli - 1,3%. The CTX-M gene, determining the resistance of Enterobacteriaceae to the 1st generation cephalosporins, was present in 3,6% of E. coli samples. The blaDHA gene, determining the resistance of Enterobacteriace to penicillins and cephalosporins of 3rd and 4th generation, was present in 34,6% of E. coli samples. In 3,6% of samples with E. адИ, two mutation genes CTX-M and blaDHA were present simultaneously. The MecA gene, which determines the resistance of S. aureus to second-generation cephalosporins, was present in 10,6% of the samples. Resistance Staphylococcus spp. and Streptococcus spp. to macrolides of the 1st generation (ErmB gene) was determined in 45,3% of samples. The conducted researches testify to a wide spread of some genes of antimicrobial resistance among pathogens in subclinical mastitis in dairy cows, which should be taken into account when choosing antibacterial therapy.
