Correlation between cytokine profile and oxidative-antioxidative status of albino rats under experimental T-2 toxicosis

Summary. The indices of lipid peroxidation – antioxidant protection system, cytokine profile and their correlation in albino rats under experimental subacute T-2 toxicosis have been studied. The research included outbred rats weighed 230-150 g. Three groups (each of 12 animals) were formed. The rats of the first group (intact) were the controls. Subacute intoxication in animals of the second and third groups was caused by daily introduction of T-2 toxin with feed during 6 days in doses of 1/20 LD50 (140 μg/kg) and 1/5 LD50 (560 μg/kg), respectively. Blood and liver con-tent of malonic dialdehyde (MDA), indices of antioxidant protection system (catalase and glutathione peroxidase activity, concentration of vitamins A and E), leukogram, content of T- and B-lymphocytes, cytokine profile were determined twenty-four hours after the last introduction of T-2 toxin in experimental and intact rats. There was stated a dose-dependent effect of MDA blood and liver accumulation in rats under T-2 toxicosis connected with catalase inhibition and decrease of non-enzymatic antioxidants blood concentration. It is shown that the intensity of lipid peroxidation (LPO) under the impact of T-2 toxin is one of the factors promoting formation of cytokine imbalance. It can be concluded that pathogenic effects of T-2 toxin in vivo are connected with the intensification of active forms of oxygen production and blood accumulation of toxic products of LPO that leads to the disorder of cytokine synthesis caused by immune cells, damage of their membranes and possibly to apoptosis. These processes are reflected by imbalance in populations of immunocytes and cytokines produced by them that has a negative impact on the organism resistance to the pathogens. The results of the research prove the need of decrease of negative impact of T-2 toxin (obtained with feeds) on animals. So, it is necessary to use sorbents, antioxidant drugs and means correcting post-intoxication disorders of immune homeostasis.
